氯硝柳胺及EDTA对不同螺龄湖北钉螺酚氧化酶活性的影响

摘要: 目的 探讨氯硝柳胺及EDTA对不同螺龄湖北钉螺酚氧化酶(PO)活性的影响。方法 将不同螺龄湖北钉螺(即成螺、幼螺、螺卵)软体匀浆、离心获取粗制酶液,再依次通过盐析、透析及凝胶过滤层析而获取部分纯化的酚氧化酶酶液。以邻苯二酚作为底物,检测不同浓度氯硝柳胺及EDTA对不同螺龄湖北钉螺PO活性的影响。结果 随着氯硝柳胺和EDTA浓度的增大,不同螺龄湖北钉螺PO活性均逐渐降低。结论 氯硝柳胺和EDTA对不同螺龄湖北钉螺PO活性均有抑制作用。

Abstract:

To investigate the effects of niclosamide and EDTA on the activity of phenoloxidase(PO) from Oncomelania hupensis in different ages. Methods The soft tissues from Oncomelania hupensis in different ages, such as adult snails, young snails and the eggs of snails, were homogenated and centrifugated to obtain crude enzyme liquid, then purified by salt precipitation, dialysis and gel filtration to obtain PO. The activity of PO was detected with catechol as the substrate in the condition of different concentrations of niclosamide and EDTA. Results The activity of PO from Oncomelania hupensis in different ages was decreasing with the concentration elevating of niclosamide and EDTA. Conclusion Both niclosamide and EDTA have inhibition effect on the activity of POfrom Oncomelania hupensis in different ages.
